Summarize

It seems impossible to find fund supporting my next year. I have to consider other ways.

I summarized my recent data. The whole picture is still not clear because the old data always conflict with the new data. I can't tell which data tell me the truth which are just the technical problem. I will discuss with my mentors about my recent data and also what I should do in the next step and next year. Several points should be very clear, becaus all my data support this.

1. Transformation is independent of antibiotics. Even without antibiotics transformation can still take place.
2. Transformation can be induced by antibitocs. That is before replica let cells grow on the plates with low concentration of antibiotics transformants appear on the second plates with high concentration of antibiotic quicker. The induction effect can be observed more obviously on plates with high concentration of spectinomycin than that on plates with high concentration of ampicillin.
3. Both of my two evidences supporting the existance of ycaI dependent pathway suffers from my recent data. Firstly, the layout of wild type transformants on ampicillin plates similar to the layout on specinomycin plates was also observed. Secondly, dprA mutant without antibiotic treatment also produce transformants 36 h later. This makes the whole picture dark again.

Although these data disapprove many of my previous suppositions. They tell me a trend of the effect of antibiotics on transformation. It seems that both transformation in ycaI mutant and dprA is seriously affected by antibiotic concentration. And the mutants' response to antibiotic concentration is different from that of the wild type. If I want to confirm this. I might have to do a lot of experiments to confirm that. And it is quite laborous to do these experiments. I need to try to find another way to approach the classical transformation.

Nevertheless, with these data I can make the conclusion that antibiotics speed up transformation in natural plasmid transformation of Escherichia coli. But the mechanism is still unkown. So I find another phenomenon but can not explain the mechanism just like another lock with out the key.

My mentors might think that only the classical transformation is more attractive because the several orders difference is more competitive and good data are more useful for publication and grant applying. I agree with him that to win the competition in the cometitive era, we have to do research which attracts as many eyes as possible to make greater impact. And since we can not explain it well at this moment since it is very difficult to continue, we have to choose other subjects which we can try to do it well. In addition, few people wish to hear a sound not to limit the using of some products. That is few enterprises will finantical support the research about proving that antibiotics is dangerous and fighting against the utilization of antibiotics but will fully support the research about developing a novel antibiotic with great market profits. This should not be the fault of researchers because we need to survive and supports from society are important to our work and life. I will also choose to do something more popular if I can not get funds to continue my study in the future professional career for surviving. But all these shoud not be the excuse. After all, it is our mission to find the truth. It is the time to reconsider the responsibility of the researchers and our society.

le chemin futur, dur ou chanceux?

Today, I discussed with Claverys about what we should do next. We will have to go back to the beginning and screen for sequences and genes important to natural transformation of E. coli, rather than choose special targets. Nevertheless, we get enough information that there is another channel through which DNA enters into cells. This is exiting although no body knows what kind of difficulties and chances we will meet in the future.

How DNA enter into E. coli??

In my experiments, it clearly shows that there is another pathway for the DNA uptake. The pathway is independent of traditional DNA uptake machine. But how does DNA enter into the cell? The question drives me mad.
